Title: Genome and Transcriptome Analysis of Clostridium cellulolyticum H10

Abstract: Clostridium cellulolyticum strain H10 (ATCC 35319) is a non-ruminal mesophilic cellulolytic bacterium which have capability of anaerobic fermentation of cellulosic plant materials, yielding acetate, ethanol, lactate, and H2, which could be used as alternative energy and commodity chemicals. Here we report the complete 4068724-bp sequence of Clostridium cellulolyticum strain H10 that comprises 3390 protein coding regions, 63 tRNA genes and 24 rRNA genes, showing comparative high G + C content (37.4%) with other clostridium species. And then we use a high-throughput sequencing-based approach (RNA-Seq) in assembling the first comprehensive, single-nucleotide resolution view of a cellulolytic bacterial transcriptome. We sample the Clostridium cellulolyticum strain H10 under three growth conditions and analysis of 31.5 million reads of sequenced cDNAs yielded an accurate and high-resolution map of transcript start sites and operon structure throughout the genome. What’s more, the sequence data identified previously nonannotated regions with significant transcriptional activity, 3'- and 5'- untranslated regions (UTR), Cis- and Trans- Encoded Base Pairing sRNAs and enhanced the accuracy of existing genome annotation.
